Brief introduction: Three consecutive stop signal sequences are inserted upstream of the Cas9D10A and EGFP elements, preventing transcription even under a CAG strong promoter. After the rats are infected by specific virus expressing Cre and target sgRNA(s), Cas9D10A protein can be expressed in specific tissues or cells and EGFP fluorescence can be observed. This approach allows specific knockout or mutation of genes. The Rosa26-LSL-Cas9D10A-EGFP sequence is knocked into one chromosome of rat heterozygotes, which are normal in growth and reproduction phenotypes.

Phenotypic characteristics: B-Cas9D10A cKI rats are effective model rats mainly applied in specific deletion of single or multiple genes in different tissues and cells. This is accomplished by infecting the model rats using a specific virus expressing Cre and target gene sgRNA(s), thereby deleting one or multiple genes.
