图片4.pngCRISPR (clustered, regularly interspaced, short palindromicrepeats) is an immunologic mechanism from bacteria that degrades invasive viral DNA or other exogenous DNA. In bacteria and archaebacteria, the CRISPR system is divided intothree types. Type I and type III systems need multiple CRISPR-related proteins (Casprotein) to play a role, while type II only needs the Cas9 protein, making it a convenient tool with wide applications. The CRISPR/Cas9 system from Streptococcus pyogenes is currently the most commonly used one.

There are two nuclease domains in the Cas9 protein that can cut the DNA double strands. The Cas9 protein first binds to the crRNA and tracrRNA to form a complex, and then binds to the target sequences to form the RNA-DNA complex structure, and finally cuts the targeted DNA double strands.

CRISPR RNA (crRNA) recognizes target DNA sequences, and tracrRNA is a conserved essential component for Cas9 activity. In order to simplify gene-editing procedures in the laboratory, crRNA and tracrRNA are fused together to make Single Guide RNAs (sgRNAs). The target gene can be edited by transfectingcells with a plasmid that expresses both the sgRNA and the Cas9 protein. The CRISPR/Cas9 technology has been successfully used in bacteria, yeast, plant, fish and mammals, and is the most efficient genome-editing technique.

The homologous recombination (HR) efficiency between exogenous DNA and target genome is very low using the standard CRISPR/ Cas9 technology. To improve HR efficiency, BIOCYTOGEN developed the Extreme Genome Editing System (EGE) which increases the HR rate 10-20 fold over standard CRISPR/Cas9 technology. This makes gene engineering with the EGE system fast and more convenient. The EGE system can edit the DNA sequence accurately at almost every genomic locus, which enables the generation of conditional knockouts, whole-gene knockouts, gene knockins, and gene point mutations. BIOCYTOGEN successfully registered the “EGE” brand in July 2015.
