  • mRNA expression analysis:
    Murine Il21r mRNA was detected in splenocytes isolated from wild-type mice, while human IL21R mRNA was detected only in humanized B-hIL21R mice.
  • Protein expression analysis:
    Murine IL21R protein was detected in wild-type mice, while human IL21R protein was exclusively detected in humanized B-hIL21R mice.
  • Leukocyte cell subpopulation analysis:
    IL21R humanization does not change the overall development, differentiation or distribution of immune cell types in the spleen, lymph node or blood.
  • Functional analysis:
    STAT3 phosphorylation was successfully induced with mouse IL21 or human IL21 in humanized B-hIL21R mice.
  • Antibody efficacy evaluation:
    Human IL21-Fc was effective in controlling tumor growth in B-hIL21R mice, demonstrating that the B-hIL21R mice provide a powerful preclinical model for in vivo evaluation of IL21 cytokine or fusion protein drugs.
