SFN 2024: A novel mouse model for in vivo evaluation of anti-human CD98HC antibody penetration across blood-brain barrier

Summary:

Protein expression analysis:

  • Mouse CD98HC protein was detectable on T cells and B cells in the spleens of WT C57BL/6 mice. In contrast, human CD98HC was exclusively detectable in homozygous B-hCD98HC mice and not in WT mice.
  • Human CD98HC was detectable on the brain microvascular endothelium of homozygous B-hCD98HC mice but not in WT C57BL/6 mice.

Leukocyte subpopulation analysis:

  • Humanization of the CD98HC gene does not affect the overall frequency or distribution of immune cell types in the spleen, blood, and lymph nodes.

Blood routine test and Blood biochemical test :

  • Humanizing CD98HC does not alter blood cell composition and morphology, nor does it affect ALT and AST levels or liver health.
