Protein Expression Analysis

Splenocytes from homozygous B-hPD-1/hCD40 mice were analyzed by flow cytometry. Mouse CD40+ and PD-1+ T cells were detectable in wild type (WT) C57BL/6, while hCD40+ and hPD-1+ T cells were detectable in the homozygous B-hPD-1/hCD40 mice.

Application

Antibody Efficacy Evaluation

B-hPD-1/hCD40 Mice

Murine colon cancer MC38 cells were subcutaneously implanted into homozygous B-hPD-1/hCD40 mice. Mice were grouped when the tumor size was approximately 150±50 mm3 (n=8). Anti-hCD40 antibody Selicrelumab Analog and the anti-hPD-1 antibody Keytruda show modest inhibition of tumor growth. On the other hand, combination of Selicrelumab Analog and Keytruda shows significantly inhibition when compared to single Ab treatment. B-hPD-1/hCD40 mouse model is a powerful tool for in vivo evaluation of combination therapy efficacy for hPD-1 antibodies and hCD40 antibodies.
