The B-CD150-iCreERT2 mice were crossed with B-CAG-tdTomato cKI mice (stock no. 110148) to obtain heterozygous B-CD150-iCreERT2; tdTomato mice. Blood cells were collected from wild-type C57BL/6 mice, heterozygous B-CAG-tdTomato cKI mice and heterozygous B-CD150-iCreERT2; tdTomato mice after 48 h i.p. injected with tamoxifen (each mouse is injected with 100μL of tamoxifen, dissolved in corn oil, at a concentration of 20 mg/mL) for five consecution days and analyzed by flow cytometry. The flow cytometry results showed that the expression of tdTomato fluorescent protein was not detected in the immune cells of wild-type mice, B-CAG-tdTomato cKI mice, and B-CD150-iCreERT2; tdTomato mice without tamoxifen induction (Fig. A and B). After tamoxifen induction, the expression of tdTomato fluorescent protein was not detected in the immune cells of wild-type and B-CAG-tdTomato cKI mice. However, the expression of tdTomato fluorescent protein was detected in the neutrophils and macrophages of anticoagulant cells in B-CD150-iCreERT2; tdTomato mice; no detection was found in unactivated B cells, T cells, NK cells, and monocytes. (Fig. C and D).
