Gene targeting strategy for B-hANGPTL4 mice. The exons 1-7 of mouse Angptl4 gene that encode the full-length protein were replaced by human ANGPTL4 exons 1-7 in B-hANGPTL4 mice.

mRNA expression analysis

Strain specific analysis of ANGPTL4 gene expression in wild-type mice and B-hANGPTL4 mice by RT-PCR. Mouse Angptl4 mRNA was detectable in fat of wild-type mice (+/+). Human ANGPTL4 mRNA was detectable in homozygous B-hANGPTL4 mice but not in wild-type mice.

Protein expression analysis

Strain specific ANGPTL4 expression analysis in homozygous B-hANGPTL4 mice by western blot. Fatty tissue was collected from wild-type mice (+/+) and homozygous B-hANGPTL4 mice (H/H), and analyzed by western blot. ANGPTL4 was both detectable in wild-type mice and B-hANGPTL4 mice (H/H) due to the cross-reactivity of antibody. And the levels of ANGPTL4 was higher under fasted state both in wild-type mice and B-hANGPTL4 mice.
