Basic Information
-
Gene targeting strategy
-
Gene targeting strategy for B-hC1Q mice. Exons 1-3 of mouse C1qa, C1qb and C1qc genes that encode the full-length protein were respectively replaced by the exons 1-3 of human C1QA, C1QB and C1QC genes in B-hC1Q mice.
-
mRNA expression analysis
-
Species-specific C1QA, C1QB and C1QC gene expression analysis in wild-type and humanized B-hC1Q mice by RT-PCR. Murine C1qa, C1qb and C1qc mRNA transcripts were detected in thymocytes isolated from wild-type C57BL/6 (+/+) mice, while human C1QA, C1QB and C1QC mRNA transcripts were detected in homozygous B-hC1Q (H/H) mice.

Protein expression analysis
-
Species-specific C1Q protein expression analysis in wild-type and humanized B-hC1Q mice. Serum was collected from wild-type C57BL/6 (+/+) and homozygous B-hC1Q (H/H) mice and analyzed by ELISA using species-specific C1Q kits. Murine C1Q protein was detected in wild-type mice, while human C1Q protein was detected in both wild-type and B-hC1Q mice, indicating that this anti-human C1Q antibody is cross-reactive between human and mouse.
