Basic Information
-
Targeting strategy
-
Gene targeting strategy for B-hCCL22 mice.
The exons 1-3 of mouse Ccl22 gene that encode the full-length protein were replaced by human CCL22 exons 1-3 in B-hCCL22 mice.
-
mRNA expression analysis
-
Strain specific analysis of CCL22 gene expression in wild type (WT) mice and B-hCCL22 mice by RT-PCR. Mouse Ccl22 mRNA was detectable only in thymus of WT mice (+/+). Human CCL22 mRNA was detectable only in homozygous B-hCCL22 mice (H/H) but not in WT mice (+/+).
-
Protein expression analysis
-
Strain specific CCL22 expression analysis in wild type (WT) mice and B-hCCL22 mice by ELISA. Thymus homogenates were collected from WT mice (+/+) and homozygous B-hCCL22 mice (H/H), and analyzed by ELISA with species-specific CCL22 ELISA kit. Mouse CCL22 was detectable in WT mice. Human CCL22 was exclusively detectable in homozygous B-hCCL22 mice (H/H) but not WT mice (+/+) . Values are expressed as mean ± SEM. ND: not detectable.
