Gene targeting strategy for B-hCD59 mice. The genomic sequences from exon 1 of mouse Cd59b to exon 5 of mouse Cd59a, which encode the full-length protein of mouse CD59B and CD59A, were replaced by exons 1-4 of the human CD59 gene in B-hCD59 mice.

CD59 Expression in Lymphocytes

Mouse CD59a and human CD59 expression analysis in T cells and B cells. T cells and B cells were isolated from the blood of wild type C57BL/6 mice, homozygous B-hCD59 mice and human blood cells. CD59 expressed on T cells and B cells were analyzed by flow cytometry with anti-CD59 antibody. Mouse CD59a were detectable on T cells and B cells of wild type C57BL/6, but not on the cells of B-hCD59 mice and human blood cells. Human CD59 was detectable on T cells and B cells of B-hCD59 mice and human blood cells, but not on the cells of wild type C57BL/6 mice.

CD59 Expression in NK, DC, and Macrophages

Mouse CD59a and human CD59 expression analysis in NK, dendritic cells and macrophages. NK cells, dendritic cells and macrophages were isolated from blood of wild type C57BL/6 mice and homozygous B-hCD59 mice. CD59 expressed on NK cells, dendritic cells and macrophages were analyzed by flow cytometry with anti-CD59 antibody. Mouse CD59a were detectable on NK cells, dendritic cells and macrophages of wild type C57BL/6, but not on the cells of B-hCD59 mice. Human CD59 was exclusively detectable on NK cells, dendritic cells and macrophages of B-hCD59 mice.

Mouse CD59a and human CD59 expression analysis in monocytes and granulocytes. Monocytes and granulocytes were isolated from the blood of wild type C57BL/6 mice, homozygous B-hCD59 mice and human blood cells. CD59 expressed on monocytes and granulocytes were analyzed by flow cytometry with anti-CD59 antibody. Mouse CD59a was detectable on monocytes and granulocytes of wild type C57BL/6 mice, but not on the cells of B-hCD59 mice and human blood cells. Human CD59 was detectable on monocytes and granulocytes of B-hCD59 mice and human blood cells, but not on the cells of wild type C57BL/6 mice.

CD59 expressed on platelets and red blood were analyzed by flow cytometry with anti-CD59 antibody. Mouse CD59a was detectable on platelets and red blood cells of wild type C57BL/6 mice, but not on the cells of B-hCD59 mice and human blood cells. Human CD59 was detectable on platelets and red blood cells of B-hCD59 mice and human blood cells, but not on the cells of wild type C57BL/6 mice.
