Gene targeting strategy for B-hIL10/hIL10RA mice. The exons 1-5 of mouse Il10 gene that encode the full-length protein were replaced by human IL10 exons 1-5 in B-hIL10/hIL10RA mice. The exons 1-6 of mouse Il10ra gene that encode the extracellular protein were replaced by human IL10RA exons 1-6 in B-hIL10/IL10RA mice.

Strain specific IL10 expression analysis in homozygous B-hIL10/hIL10RA mice by ELISA. Serum was collected from wild-type C57BL/6 mice (+/+) and homozygous B-hIL10/hIL10RA mice (H/H) stimulated with LPS in vivo, and analyzed by ELISA with species-specific IL10 ELISA kit. Mouse IL10 was only detectable in wild-type mice. Human IL10 was exclusively detectable in homozygous B-hIL10/hIL10RA mice but not wild-type mice.
Strain specific IL10RA expression analysis in homozygous B-hIL10/hIL10RA mice by flow cytometry. Peritoneal exudative macrophages were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hIL10/hIL10RA mice (H/H), and analyzed by flow cytometry with species-specific anti-IL10RA antibody. Mouse IL10RA was detectable in wild-type mice. Human IL10RA were exclusively detectable in homozygous B-hIL10/hIL10RA mice but not in wild-type mice.
