Gene targeting strategy for B-hIL3 mice. Exons 1-5 of the mouse Il3 gene that encodes the full-length protein were replaced by human IL3 exons 1-5 in B-hIL3 mice.

mRNA expression analysis

Species-specific IL3 gene expression analysis in wild-type and humanized B-hIL3 mice by RT-PCR. Mouse splenocytes were isolated and stimulated with ConA for 48 hrs in vitro. Mouse Il3 mRNA was detected in wild-type C57BL/6 (+/+) mice, while human IL3 mRNA was detected in homozygous B-hIL3 (H/H) mice.

Species-specific IL3 expression analysis in wild-type and humanized B-hIL3 mice. Splenocyte culture supernatant was collected from wild-type C57BL/6 (+/+) and homozygous B-hIL3 (H/H) mice, stimulated with PMA/ionomycin in vitro, and analyzed by ELISA using species-specific IL3 ELISA kits. Mouse IL3 was detected in wild-type mice, while human IL3 was detected in B-hIL3 mice.
