Basic Information
-
Targeting strategy
-
Gene targeting strategy for B-hIL36A mice.
The exon 2-5 of mouse Il36a gene that encodes the full-length protein were replaced by human IL36A exon 1-4 in B-hIL36A mice.
-
mRNA expression analysis
-
Strain specific analysis of IL36A gene expression in wild type (WT) mice and B-hIL36A mice by RT-PCR.
Mouse Il36a mRNA was detectable only in stomach of WT mice (+/+). Human IL36A mRNA was detectable only in homozygous B-hIL36A mice (H/H) but not in WT mice (+/+).
-
Protein expression analysis
-
Strain specific IL36A expression analysis in homozygous B-hIL36A mice by western blot .
Skin were collected from wild type (WT) mice (+/+) and homozygous B-hIL36A mice (H/H), and analyzed by western blot with anti-IL36A antibody. Mouse IL36A was detectable in WT mice (+/+) due to the cross-reactivity of antibodies. Human IL36A was detectable in homozygous B-hIL36A mice (H/H) but not in WT mice (+/+).

Summary
-

Mouse Il36a mRNA was detectable only in stomach of WT mice (+/+). Human IL36A mRNA was detectable only in homozygous B-hIL36A mice (H/H) but not in WT mice (+/+).
Protein expression analysis:
Human IL36A was detectable in skin of homozygous B-hIL36A mice (H/H) but not in WT mice (+/+), and mouse IL36A was detectable in WT mice (+/+) due to the cross-reactivity of antibodies.
