Gene targeting strategy for B-hTGM2 mice. The exons 1-13 of mouse Tgm2 gene that encode the full coding sequences were replaced by human TGM2 exons 1-13 in B-hTGM2 mice.

mRNA expression analysis

Species-specific TGM2 gene expression analysis in wild-type and humanized B-hTGM2 mice by RT-PCR. Murine Tgm2 mRNA was detected in ovarian tissue isolated from wild-type C57BL/6 (+/+) mice but not in homozygous B-hTGM2 mice (H/H). Human TGM2 mRNA was detectable only in homozygous B-hTGM2 mice but not in wild-type mice.

Protein expression analysis

TGM2 protein expression analysis in humanized B-hTGM2 mice. Liver, lung and spleen tissues were isolated from wild-type C57BL/6 (+/+) and homozygous B-hTGM2 (H/H) mice and analyzed by western blot using an anti-m/hTGM2 antibody. HeLa cells were used as a positive control for human TGM2 protein expression. TGM2 protein was detected in wild-type and B-hTGM2 mice with a known cross-reactive anti-TGM2 antibody.
