Gene targeting strategy for B-hWNT3A mice. The mouse Wnt3a gene that encode the extracellular domain were replaced by human WNT3A counterpart gene in B-hWNT3A mice.

Strain specific analysis of WNT3A gene expression in wild-type mice and B-hWNT3A mice by RT-PCR. Mouse Wnt3a mRNA was detectable in lung of wild-type mice (+/+). Human WNT3A mRNA was detectable only in homozygous B-hWNT3A mice (H/H) but not in wild-type mice.
Strain specific WNT3A expression analysis in homozygous B-hWNT3A mice by western blot. Heart tissue and lung tissue were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hWNT3A mice (H/H), and analyzed by western blot with anti-hWNT3A antibody. Human WNT3A was detectable in wild-type mice and homozygous B-hWNT3A mice, as the antibody is crossly reactive with WNT3A in human and mice.
