mRNA expression analysis

Strain specific analysis of CCR4 gene expression in WT and hCCR4 mice by RT-PCR. Mouse Ccr4 mRNA was detectable only in splenocytes of wild-type (+/+) mice. Human CCR4 mRNA was detectable only in H/H, but not in +/+ mice.

Strain specific CCR4 expression analysis in homozygous B-hCCR4 mice by flow cytometry. Splenocytes were collected from WT and homozygous B-hCCR4 (H/H) mice, and analyzed by flow cytometry with species-specific anti-CCR4 antibody. Mouse CCR4 was detectable in WT mice. Human CCR4 was exclusively detectable in homozygous B-hCCR4 but not WT mice.

Analysis of B-CAG-hCCR4 MC38 by FACS. Flow cytometry analysis of the B-CAG-hCCR4 MC38 was performed to assess anti-human CCR4 Ab binding. Single live cells were gated for CD45- population and used for further analysis as indicated here. B-CAG-hCCR4 MC38 can bind well to anti-hCCR4 antibody (mogamulizumab, in house) vs isotype control.
