The exon 2 of mouse Cxcr6 gene that encode the extracellular domain were replaced by human CXCR6 exon 2 in B-hCXCR6 mice.

Protein expression analysis in NK cells

Strain specific CXCR6 expression analysis in homozygous B-hCXCR6 mice by flow cytometry. Splenocytes were collected from wild-type mice (+/+) and homozygous B-hCXCR6 mice (H/H), and analyzed by flow cytometry with species-specific anti-CXCR6 antibody. Mouse CXCR6 was detectable in wild-type mice. Human CXCR6 was exclusively detectable in homozygous B-hCXCR6 but not in wild-type mice.

Species-specific CXCR6 protein expression analysis in wild-type and humanized B-hCXCR6 mice. Splenocytes were isolated from wild-type C57BL/6 (+/+) and homozygous B-hCXCR6 (H/H) mice and analyzed by flow cytometry using species-specific anti-CXCR6 antibodies. Murine CXCR6 protein was detected in wild-type mice, while human CXCR6 protein was detected in B-hCXCR6 mice.

Human CXCR6 was both detectable on NK cells and NKT cells in homozygous B-hCXCR6 mice but not in wild-type mice, and mouse CXCR6 was detectable only in wild-type mice.
