Gene targeting strategy for B-hADAM15 mice.
The exons 1-17 of mouse Adam15 gene that encode the extracellular domain were replaced by human ADAM15 exons 1-17 in B-hADAM15 mice. -
Protein expression analysis

Strain specific ADAM15 expression analysis in C57BL/6 mice and homozygous B-hADAM15 mice by western blot. Various tissue lysates were collected from wild-type C57BL/6 mice(+/+) and homozygous B-hADAM15 mice(H/H), and then analyzed by western blot with anti-ADAM15 antibody. 40 μg total proteins were loaded for western blotting analysis.

mRNA expression analysis

Strain specific analysis of ADAM15 mRNA expression in wild-type C57BL/6 mice and B-hADAM15 mice by RT-PCR. Liver RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hADAM15 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human ADAM15 primers. Mouse Adam15 mRNA was detectable only in wild-type C57BL/6 mice. Human ADAM15 mRNA was detectable only in homozygous B-hADAM15 mice but not in wild-type mice.
