Targeting strategy

Gene targeting strategy for B-hB2M/HLA-G mice. The exons 1-3 of mouse B2m gene was replaced by human B2M and HLA-G gene in B-hB2M/HLA-G mice.

Protein expression analysis

Strain specific hB2M expression analysis in heterozygous B-hB2M/HLA-G (H/+) mice by flow cytometry. Splenocytes were collected from wild-type mice (+/+) and heterozygous B-hB2M/HLA-G (H/+) mice, and analyzed by flow cytometry with species-specific anti-B2M antibody. Mouse B2M was detectable in wild-type mice and heterozygous B-hB2M/HLA-G (H/+). Human B2M was exclusively detectable in heterozygous B-hB2M/HLA-G (H/+) mice but not wild-type mice.
