Gene targeting strategy for B-hCD160 mice. The exons 2-5 of mouse Cd160 gene that encode the full-length region, including signal peptide were replaced by human CD160 exons 3-6 in B-hCD160 mice.

mRNA expression analysis

Strain specific analysis of CD160 gene expression in wild-type mice and B-hCD160 mice by RT-PCR. Mouse Cd160 mRNA was detectable in splenocytes of wild-type mice (+/+). Human CD160 mRNA was detectable only in homozygous B-hCD160 mice (H/H) but not in wild-type mice.

Protein expression analysis

Strain specific CD160 expression analysis in homozygous B-hCD160 mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCD160 mice (H/H) which sitimulated with the serial dilutions of mouse IL-2 for 18h, and analyzed by flow cytometry with species-specific anti-CD160 antibody. Mouse CD160 was detectable in wild-type mice. Human CD160 was exclusively detectable in homozygous B-hCD160 mice but not in wild-type mice.

Human CD160 was detectable on T cells, B cells and NK cells in homozygous B-hCD160 mice but not in wild-type mice, and mouse CD160 was detectable only in wild-type mice.
