Gene targeting strategy for B-hCD70 mice. The exons 1-3 of the mouse Cd70 gene that encode the extracellular domain were replaced by human CD70 exons 1-3 in B-hCD70 mice.

mRNA expression analysis

Strain specific analysis of CD70 gene expression in wild type (WT) mice and B-hCD70 mice by RT-PCR. Mouse CD70 mRNA was detectable only in thymocytes of WT mice (+/+) and B-hCD70 mice (H/+). Human CD70 mRNA was detectable only in homozygous B-hCD70 mice (H/+) but not in WT mice (+/+).

Protein expression analysis

Species-specific CD70 protein expression analysis in humanized B-hCD70 mice. Following LPS stimulation in vivo, bone marrow-derived dendritic cells were isolated from wild-type C57BL/6 (+/+) and homozygous B-hCD70 (H/H) mice and analyzed by flow cytometry using species-specific anti-CD70 antibodies. Murine CD70 protein was detected in wild-type mice, while human CD70 protein was detected in B-hCD70 mice.
