The exons 1-5 of mouse CD80 gene that encode the extracellular domain were replaced by human CD80 exons 2-5 in B-hCD80 mice.

Strain specific analysis of CD80 gene expression in wild type (WT) mice and B-hCD80 mice by RT-PCR.
Mouse Cd80 mRNA was detectable only in splenocytes of WT mice (+/+). Human CD80 mRNA was detectable only in homozygous B-hCD80 mice (H/H) but not in WT mice (+/+).

Peritoneal exudative macrophages (PEMs) were collected from wild type (WT) mice (+/+) and homozygous B-hCD80 mice (H/H), and analyzed by flow cytometry with species-specific anti-CD80 antibody. Mouse CD80 was detectable in WT mice (+/+). Human CD80 was exclusively detectable in homozygous B-hCD80 mice (H/H) but not in WT mice (+/+).
