Gene targeting strategy for B-hCLEC14A mice. The exon 1 of mouse Clec14a gene that encodes the extracellular domain was replaced by human CLEC14A exon 1 in B-hCLEC14A mice.

mRNA and protein expression analysis

Strain specific analysis of CLEC14A gene expression in wild-type C57BL/6 mice and hCLEC14A mice by RT-PCR and western blot. (A) mRNA expression. Mouse Clec14a mRNA was detectable only in lung of wild-type C57BL/6 mice (+/+). Human CLEC14A mRNA was detectable only in homozygous B-hCLEC14A mice (H/H), but not in wild-type mice. (B) Protein expression. Kidney was collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCLEC14A mice (H/H), and analyzed by western blot with a known cross-reactive anti-CLEC14A antibody. The CLEC14A protein can be detected in homozygous B-hCLEC14A mice.
