Gene targeting strategy for B-hEPCAM mice. The exons 3-8 of mouse Epcam genes that encode the extracellular protein were replaced by human EPCAM exons 2-7 in B-hEPCAM mice.

Protein expression analysis

Strain specific EPCAM expression analysis in homozygous B-hEPCAM mice by flow cytometry. Lung, small intestine and pancreas were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hEPCAM mice (H/H), and CD45- cells were analyzed by flow cytometry with species-specific anti-EPCAM antibody. Mouse EPCAM was detectable in wild-type mice. Human EPCAM was exclusively detectable in homozygous B-hEPCAM (H/H) but not in wild-type mice.
