Gene targeting strategy

Gene targeting strategy for B-hICAM1 mice. The exons 2-7 of mouse Icam1 that encode the full-length protein, not including signal peptides were replaced by the exons 2-7 of human ICAM1 gene in B-hICAM1 mice.

Protein expression analysis of ICAM1 in T cells and B cells

Strain specific ICAM1 expression analysis in homozygous B-hICAM1 mice by flow cytometry.

T cells and B cells from spleen were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hICAM1 mice (H/H), and analyzed by flow cytometry with species-specific anti-ICAM1 antibody. Mouse ICAM1 was detectable in wild-type mice. Human ICAM1 was exclusively detectable in homozygous B-hICAM1 mice but not in wild-type mice.

Protein expression analysis of ICAM1 in NK cells, dendritic cells

Strain specific ICAM1 expression analysis in homozygous B-hICAM1 mice by flow cytometry.

NK cells, dendritic cells from spleen were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hICAM1 mice (H/H), and analyzed by flow cytometry with species-specific anti-ICAM1 antibody. Mouse ICAM1 was detectable in wild-type mice. Human ICAM1 was exclusively detectable in homozygous B-hICAM1 mice but not in wild-type mice.

Protein expression analysis of ICAM1 in granulocytes, monocytes and macrophages

Strain specific ICAM1 expression analysis in homozygous B-hICAM1 mice by flow cytometry.

Granulocytes, monocytes and macrophages from spleen were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hICAM1 mice (H/H), and analyzed by flow cytometry with species-specific anti-ICAM1 antibody. Mouse ICAM1 was detectable in wild-type mice. Human ICAM1 was exclusively detectable in homozygous B-hICAM1 mice but not in wild-type mice.

Summary

Protein expression analysis:

Human ICAM1 was detectable on T cells, B cells, NK cells, dendritic cells, granulocytes, monocytes, macrophages in homozygous B-hICAM1 mice but not in wild-type mice, and mouse ICAM1 was detectable only in wild-type mice.
