Gene targeting strategy for B-hITGA5 mice. The exons 1-29 of mouse Itga5 gene that encode the extracellular domain were replaced by human ITGA5 exons 1-29 in B-hITGA5 mice.

Species-specific ITGA5 gene expression analysis in wild-type and humanized B-hITGA5 mice by RT-PCR. Murine Itga5 mRNA was detected in bladder tissue isolated from wild-type (+/+) mice, while human ITGA5 mRNA was detected in homozygous B-hITGA5 (H/H) mice.

Species-specific ITGA5 protein expression analysis in wild-type and humanized B-hITGA5 mice. Following anti-CD3e stimulation in vivo (7.5 μg/mice, 24 hours, i.p.), splenocytes were isolated from wild-type C57BL/6 (+/+) and homozygous B-hITGA5 (H/H) mice and analyzed by flow cytometry using species-specific anti-ITGA5 antibodies. Murine ITGA5 protein was detected in wild-type mice, while human ITGA5 protein was detected in B-hITGA5 mice.
