Basic Information
-
Targeting strategy
-
Gene targeting strategy for B-hNOTCH1 mice. The exons 1~28 of mouse Notch1 gene that encode the extracellular domain were replaced by human NOTCH1 exons 1~28 in B-hNOTCH1 mice.
-
Protein expression analysis in thymus
-
Strain specific NOTCH1 expression analysis in homozygous B-hNOTCH1 mice by flow cytometry. Thymus were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hNOTCH1 mice (H/H), and analyzed by flow cytometry with species-specific anti-NOTCH1 antibody. Mouse NOTCH1 was detectable in wild-type mice. Human NOTCH1 was exclusively detectable in homozygous B-hNOTCH1 mice.
-
Protein expression analysis in lung
-
Strain specific NOTCH1 expression analysis in homozygous B-hNOTCH1 mice by flow cytometry. Lung were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hNOTCH1 mice (H/H), and analyzed by flow cytometry with species-specific anti-NOTCH1 antibody. Mouse NOTCH1 was not detectable in wild-type mice. Human NOTCH1 was not detectable in homozygous B-hNOTCH1 mice.
-
Protein expression analysis in kidney
-
Strain specific NOTCH1 expression analysis in homozygous B-hNOTCH1 mice by flow cytometry. Kidney were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hNOTCH1 mice (H/H), and analyzed by flow cytometry with species-specific anti-NOTCH1 antibody. Mouse NOTCH1 was not detectable in wild-type mice. Human NOTCH1 was not detectable in homozygous B-hNOTCH1 mice.

Summary
-
Protein expression analysis:
Human NOTCH1 was detectable on epithelial cells in thymus of homozygous B-hNOTCH1 mice, and mouse NOTCH1 was detectable only in wild-type C57BL/6 mice.
