Basic Information

Targeting strategy
-
Gene targeting strategy for B-hNRP1 mice.
The exons 1-16 of mouse Nrp1 gene that encode the extracellular domain were replaced by human NRP1 exons 1-16 in B-hNRP1 mice.
-
Protein expression analysis in Treg cells
-
Strain specific NRP1 expression analysis in homozygous B-hNRP1 mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hNRP1 mice (H/H), and analyzed by flow cytometry with species-specific anti-NRP1 antibody. Mouse NRP1 was detectable in wild-type mice. Human NRP1 was exclusively detectable in homozygous B-hNRP1 but not in wild-type mice.

Protein expression analysis in CD4+ T cells

Summary
-
Protein expression analysis:
Human NRP1 was both detectable on CD4+ T cells and Treg cells in homozygous B-hNRP1 mice but not in wild-type mice, and mouse NRP1 was detectable only in wild-type mice.
