Gene targeting strategy for B-hPD-1/hBTLA mice. The exon 2 of mouse Pd-1 gene that encodes the extracellular domain was replaced by human PD-1 exon 2 in B-hPD-1/hBTLA mice. The exon 2 of mouse Btla gene that encodes the extracellular domain was replaced by human BTLA exon 2 in B-hPD-1/hBTLA mice.

Phenotype
mRNA expression analysis
Strain specific analysis of PD-1 and BTLA gene expression in WT and B-hPD-1/hBTLA mice by RT-PCR. Mouse Pd-1 and Btla mRNA was detectable only in splenocytes of wild-type (+/+) mice. Human PD-1 and BTLA mRNA was detectable only in H/H, but not in +/+ mice.
Protein Expression Analysis
Splenocytes from both wild type (WT) C57BL/6 and homozygous B-hPD-1/hBTLA mice were analyzed by flow cytometry. Mouse BTLA+ B cells were detectable in the WT C57BL/6 mice, while human BTLA+ B cells were detectable in the homozygous B-hPD-1/hBTLA mice.
Splenocytes from both wild type (WT) C57BL/6 and homozygous B-hPD-1/hBTLA mice were analyzed by flow cytometry. Mouse PD-1+ T cells were detectable in the WT C57BL/6 mice, while human PD-1+ T cells were detectable in the homozygous B-hPD-1/hBTLA mice.
