Gene targeting strategy for B-hSEMA4D mice.
The chimeric SEMA4D CDS (replace the extracellular domain) was inserted in the exon 4 of mouse Sema4d gene in B-hSEMA4D mice.

Strain specific SEMA4D expression analysis in T cells of homozygous B-hSEMA4D mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hSEMA4D mice (H/H), and analyzed by flow cytometry with species-specific anti-SEMA4D antibody. Mouse SEMA4D was detectable in wild-type mice. Human SEMA4D was exclusively detectable in homozygous B-hSEMA4D but not in wild-type mice.
