Gene targeting strategy for B-hTIM4/hTIM1 mice.
The mouse Tim4 gene that encodes the extracellular and transmembrane regions were replaced by human TIM4 counterpart in B-hTIM4/hTIM1 mice.
The mouse Tim1 gene that encode the extracellular domain were replaced by human TIM1 counterpart gene in B-hTIM4/hTIM1 mice.

Strain specific TIM4 expression analysis in homozygous B-hTIM4/hTIM1 mice by flow cytometry. Macrophages in peritoneal lavage fluid were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTIM4/hTIM1 mice (H/H), and analyzed by flow cytometry with species-specific anti-TIM4 antibody. Mouse TIM4 was detectable in wild-type mice. Human TIM4 was exclusively detectable in homozygous B-hTIM4/hTIM1 mice but not in wild-type mice.
Strain specific TIM1 expression analysis in homozygous B-hTIM4/hTIM1 mice by flow cytometry. Kidney tissue was collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTIM4/hTIM1 mice (H/H) stimulated with folic acid in vivo (5000 μg/mice, stimulation for 22 hours, i.p. ), and analyzed by flow cytometry with species-specific anti-TIM1 antibody. Mouse TIM1 was detectable in wild-type mice. Human TIM1 were exclusively detectable in homozygous B-hTIM4/hTIM1 mice but not in wild-type mice.
