Description

This B-hPD-L1 Raji-luc cell line expresses firefly luciferase as a marker of Raji cells. Fluorescence and luminescence can be observed in B-hPD-L1 Raji-luc cells.

Targeting strategy

Gene targeting strategy for B-hPD-L1 Raji-luc cells. The exogenous promoter, human PD-L1 coding sequence and luciferase was inserted into the AAVS1 locus in B-luc-GFP Raji cells.

Protein Expression Analysis

PD-L1 expression analysis in B-hPD-L1 Raji-luc cells by flow cytometry. Single cell suspensions from B-luc-GFP Raji and B-hPD-L1 Raji-luc cultures were stained with species-specific anti-PD-L1 antibody. Human PD-L1 was detected on the surface of B-hPD-L1 Raji-luc cells but not B-luc-GFP Raji cells. The 1-H11 clone of B-hPD-L1 Raji-luc cells was used for in vivo experiments.

In vitro luciferase activity of  B-hPD-L1 Raji-luc cells

Luminescence signal intensity of B-hPD-L1 Raji-luc cells. Luminescence intensity was measured using the Bright-GloTM luciferase Assay (Promega, Cat E2610). B-hPD-L1 Raji-luc cells have a strong luminescence signal. Clone No. :1-H11.

Quantitative imaging data & Body weight changes

Tumor growth and in vivo imaging of B-hPD-L1 Raji-luc cells. B-luc-GFP Raji cells and B-hPD-L1 Raji-luc cells (5×105) were injected into the tail vein of wild-type B-NDG mice. Signal intensity and body weight were measured twice a week. (A) Imaging was performed twice a week. (B) Body weight (Mean ± SEM). B-hPD-L1 Raji-luc cells can be used for in vivo efficacy evaluation.
