Description

  • APOA1 is the major protein component of high density lipoprotein (HDL) in the plasma, which is synthesized in the liver and small intestine.
  • The exons 1~4 of mouse Apoa1 gene that encodes the full-length protein was replaced by human APAO1 exons 1~4 in B-hAPOA1 mice.
  • hAPOA1 protein was detected in plasma of homozygous B-hAPOA1 mice
  • Application: This product is used as the metabolic disease models associated with APOA1 including atherosclerosis, and hypercholesterolemia.

Targeting strategy

Gene targeting strategy for B-hAPOA1 mice. The exons 1~4 of mouse Apoa1 gene that encodes the full-length protein was replaced by human APAO1 exons 1~4 in B-hAPOA1 mice.

mRNA expression analysis

Strain specific analysis of APOA1 mRNA expression in wild-type C57BL/6 mice and B-hAPOA1 mice by RT-PCR. Liver RNA was isolated from wildtype C57BL/6 mice (+/+) and homozygous B-hAPOA1 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human APOA1 primers. Mouse Apoa1 mRNA was detectable only in wild-type C57BL/6 mice. Human APOA1 mRNA was detectable only in homozygous B-hAPOA1 mice but not in wild-type mice.

Protein expression analysis in plasma

Strain specific APOA1 expression analysis in homozygous B-hAPOA1 mice by ELISA. Plasma collected from wild-type C57BL/6J mice (+/+) and homozygous B-hAPOA1 mice (H/H) were analyzed by ELISA with species-specific APOA1 ELISA kit. Human APOA1 was exclusively detectable in homozygous B-hAPOA1 mice but not in wild-type mice.
