Gene targeting strategy for B-hITGA6 mice. Exons 2-23 of the mouse Itga6 gene that encodes the extracellular domain were replaced by human ITGA6 exons 2-23 in B-hITGA6 mice.

mRNA and protein expression analysis

Strain specific analysis of ITGA6 gene expression in wild-type mice and B-hITGA6 mice by RT-PCR. Mouse Itga6 mRNA was detectable in colon of wild-type mice (+/+). Human ITGA6 mRNA was detectable only in homozygous B-hITGA6 but not in wild-type mice.
ITGA6 expression analysis in C57BL/6 wild-type mice and homozygous B-hITGA6 mice by flow cytometry. Blood platelets were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hITGA6 mice (H/H) , and analyzed with anti-ITGA6 antibody. ITGA6 was detectable in wild type mice and homozygous B-hITGA6 mice, as the antibody is crossly reactive with ITGA6 in human and mice.
