Gene targeting strategy for B-hLRRC15 mice.
The exon 2 of mouse Lrrc15 gene that encodes the extracellular region was replaced by human LRRC15 exon 2 in B-hLRRC15 mice.
mRNA and protein expression analysis
Strain specific analysis of LRRC15 gene expression in wild-type C57BL/6 mice and B-hLRRC15 mice by RT-PCR and western blot. (A) mRNA expression. Mouse Lrrc15 mRNA was detectable only in brain of wild-type C57BL/6 mice (+/+). Human LRRC15 mRNA was detectable only in homozygous B-hLRRC15 mice (H/H), but not in wild-type mice. (B) Protein expression. Brain was collected from wild-type C57BL/6 mice (+/+) and homozygous B-hLRRC15 mice (H/H), and analyzed by western blot with a known cross-reactive anti-LRRC15 antibody. Human neuroblastoma cell line SH-SY-5Y was used as positive control. The LRRC15 protein can be detected in homozygous B-hLRRC15 mice.
