Description

CD110 (MPL) is a type I transmembrane glycoprotein which functions as a receptor for thrombopoietin; binding causes the proliferation and differentiation of megakaroyocytes in addition to the production of platelets and protection of stem cells from apoptosis. CD110 is primarily expressed on hematopoietic stem cells, hematopoietic precursor cells, cells of the megakaryocytic lineage, and platelets.

Targeting strategy

Gene targeting strategy for B-hMPL mice. The exons 1-10 of mouse Mpl gene that encode extracellular domain is replaced by human counterparts in B-hMPL mice. The genomic region of mouse Mpl gene that encodes cytoplasmic portion is retained. The promoter, 5’UTR and 3’UTR region of the mouse gene are retained. The chimeric MPL expression is driven by endogenous mouse Mpl promoter, while mouse Mpl gene transcription and translation will be disrupted.

mRNA expression analysis

Strain specific analysis of MPL mRNA expression in wild-type C57BL/6JNifdc mice and B-hMPL mice by RT-PCR. Spleen RNA were isolated from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hMPL mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human MPL primers. Mouse Mpl mRNA was detectable in wild-type C57BL/6JNifdc mice. Human MPL mRNA was detectable only in homozygous B-hMPL mice (H/H) but not in wild-type mice.

Protein expression analysis

Strain specific CD110/MPL expression analysis in B-hMPL mice by flow cytometry. Blood and bone marrow were collected from wild-type C57BL/6JNifdc mice (+/+) and B-hMPL mice (H/H), and analyzed by flow cytometry with species-specific anti-human CD110/MPL antibody (BioLegend, 393803). Human CD110/MPL was exclusively detectable in homozygous B-hMPL mice but not in wild-type mice.
