Gene targeting strategy for B-hPLAT mice. The exons 2-14 of mouse Plat gene that encode the whole molecule are replaced by human counterparts in B-hPLAT mice. The promoter and 5’UTR region of the mouse gene are retained, but the 3’UTR region of the mouse gene are replaced by human counterparts. 

Strain specific PLAT expression analysis in wild-type C57BL/6JNifdc mice and homozygous B-hPLAT mice by ELISA. Serum was collected from female wild-type C57BL/6 mice (+/+) (n=3, 5-week-old) and homozygous B-hPLAT mice (H/H) (n=3, 5-week-old). Expression level of mouse and human PLAT were analyzed by ELISA (anti-mouse PLAT ELISA kit: Abcam, ab233615; anti-human PLAT ELISA kit: Abcam, ab282306). Mouse PLAT was only detectable in wild-type C57BL/6 mice. Human PLAT was exclusively detectable in homozygous B-hPLAT mice (n=3). Values are expressed as mean ± SEM.
