Basic Information
-
Targeting strategy
-
Gene targeting strategy for B-hUPA mice. The full coding region of mouse Upa was replaced by human UPA coding region sequences in B-hUPA mice.
-
mRNA expression analysis
-
Species-specific PLAU expression analysis in wild-type and humanized B-hPLAU mice by RT-PCR and western blot. (A) Murine Plau mRNA was detected in kidney tissue isolated from wild-type (+/+) mice, while human PLAU mRNA was exclusively detected in homozygous B-hPLAU (H/H) mice. (B) Liver and kidney tissues were isolated from wild-type C57BL/6 (+/+) and homozygous B-hPLAU (H/H) mice, and analyzed by western blot using an anti-m/hPLAU antibody. PLAU protein was detected in wild-type and B-hPLAU mice due to the cross-reactive antibody.

Summary
-
- mRNA expression analysis:
Human UPA mRNA was detectable only in homozygous B-hUPA mice but not in wild-type mice.
- Protein expression analysis:
UPA protein was detectable in wild-type C57BL/6 mice (+/+) and homozygous B-hUPA mice (H/H), as the antibody is crossly reactive with both human and mouse UPA.
