Gene targeting strategy for B-hSTAB1 mice. The exons 2-67 of mouse Stab1 gene that extracellular domain are replaced by human counterparts in B-hSTAB1 mice. The genomic region of mouse Stab1 gene that encodes cytoplasmic portion is retained. The encode signal peptide, transmembrane domain, promoter, 5’UTR and 3’UTR region of the mouse gene are retained. The human STAB1 expression is driven by endogenous mouse Stab1 promoter, while mouse Stab1 gene transcription and translation will be disrupted.

mRNA expression analysis

Strain specific analysis of STAB1 mRNA expression in wild-type C57BL/6JNifdc mice and B-hSTAB1 mice by RT-PCR. Brain RNA were isolated from wild-type C57BL/6JNifdc (+/+) and homozygous B-hSTAB1 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse and human STAB1 primers. Mouse Stab1 mRNA was only detectable in wild-type mice but not in homozygous B-hSTAB1 mice. Human STAB1 mRNA was only detectable in homozygous B-hSTAB1 mice but not in wild-type mice.
