Gene targeting strategy for B-hTLR3 mice.
The exons 4~6 of mouse Tlr3 gene that encode the extracellular domain were replaced by human TLR3 exons 2~4 in B-hTLR3 mice.

Strain specific TLR3 expression analysis in homozygous B-hTLR3 mice by flow cytometry. Peritoneal washes and Splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTLR3 mice (H/H). Flow cytometry analysis of the Peritoneal washes-macrophages, Splenocytes-macrophages, DCs were analyzed with anti-TLR3 antibody. Mouse TLR3 was detected in Peritoneal washes-macrophages, Splenocytes-macrophages, DCs of wild-type mice. Human TLR3 was detected in Peritoneal washes-macrophages, Splenocytes-macrophages, DCs of homozygous B-hTLR3 mice.
Strain specific TLR3 expression analysis in homozygous B-hTLR3 mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTLR3 mice (H/H). Flow cytometry analysis of the monocytes, granulocytes, NK cells were analyzed with anti-TLR3 antibody. Mouse TLR3 was detected in monocytes, granulocytes, NK cells of wild-type mice. Human TLR3 was detected in monocytes, NK cells of homozygous B-hTLR3 mice, but not in granulocytes.
