- Expression: TM4SF1 is a protein with four transmembrane domains, which is highly expressed on lung, breast, colon, and ovarian carcinomas. It is also present on normal endothelial cells.
- Application: This mice model can be used for pharmacodynamics and safety evaluation of colon, breast and other cancers.

Targeting strategy

Gene targeting strategy for B-hTM4SF1 mice. The exons 2-6 of mouse Tm4sf1 gene that encode signal peptide, extracellular domain, transmembrane domain, cytoplasmic region and 3’UTR are replaced by human counterparts in B-hTM4SF1 mice. The promoter and 5’UTR region of the mouse gene are retained. The human TM4SF1 expression is driven by endogenous mouse Tm4sf1 promoter, while mouse Tm4sf1 gene transcription and translation will be disrupted.

mRNA expression analysis

Strain specific analysis of TM4SF1 mRNA expression in wild-type C57BL/6 mice and B-hTM4SF1 mice by RT-PCR. Lung RNA was isolated from wildtype C57BL/6 mice (+/+) and homozygous B-hTM4SF1 mice (H/H), then cDNA libraries was synthesized by reverse transcription, followed by PCR with mouse or human TM4SF1 primers. Mouse Tm4sf1 mRNA was only detectable in wild-type mice. Human TM4SF1 mRNA was exclusively detectable in homozygous B-hTM4SF1 mice but not in wild-type mice.

Protein expression analysis

Strain specific TM4SF1 expression analysis in homozygous B-hTM4SF1 mice by flow cytometry. Lung endothelial cells were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTM4SF1 mice (H/H), and analyzed by flow cytometry with anti-humanTM4SF1 antibody (R&D, FAB8164R). hTM4SF1 was detectable on lung endothelial cells of homozygous mice.
