Description
- Thy-1 as a marker for thymus-derived lymphocytes.
- The locus for murine Thy-1 maps to chromosome 9 and includes two alleles termed Thy-1a and Thy-1b, which code for Thy-1.1 and Thy-1.2 glycoproteins, respectively.
- Thy-1.2 is expressed by most mouse strains, whereas Thy-1.1 is an alloantigen of the AKR/J and PL strains.
Targeting Strategy
Gene targeting strategy for B-Thy1.1 mice.
Introduce a glutamine to argnine acid mutation (Q108R) in mouse Thy1 gene, resulting that the encoding protein can be recognized by anti-Thy1.1 antibodies.
Thy1.1 expression analysis in B-Thy1.1 mice
Thy1.1 expression analysis in homozygous B-Thy1.1 mice by flow cytometry. Splenocytes, blood and thymocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-Thy1.1 mice (Mut/Mut), and then analyzed by flow cytometry with anti-Thy1.1 and anti-Thy1.2 antibodies. Thy1.2 was detectable in C57BL/6 mice. Thy1.1 were only detectable in B-Thy1.1 mice but not in wild-type mice.
