    Introduction

    Orthotopic tumor models involve implanting tumor cell lines or patient-derived xenografts orthotopically into the corresponding organ of origin in mice. This preclinical approach allows tumor growth and metastasis to develop within a physiologically relevant microenvironment, enabling precise evaluation of tumor–host interactions, immune responses, and therapeutic efficacy. Compared with subcutaneous xenograft models, orthotopic tumor models more accurately recapitulate disease progression and metastatic patterns observed in human cancers, making them invaluable tools for oncology and immuno-oncology drug development.
