Application

B-hPSMA MC38 cells have the capability to establish tumors in vivo, which can be used for efficacy studies.

Gene Targeting Strategy

Gene targeting strategy for B-hPSMA MC38 cells. The exogenous promoter and human PSMA coding sequence was inserted into the murine PSMA locus. This insertion disrupts the endogenous murine PSMA gene, resulting in a non-functional transcript.

Protein Expression Analysis

Pre-Inoculation

PSMA expression analysis in B-hPSMA MC38 cells. Single cell suspensions from B-hPSMA MC38 cultures were stained with an anti-human PSMA antibody and analyzed by flow cytometry. Human PSMA protein was detected on the surface of B-hPSMA MC38 cells compared to wild-type MC38 cells. The 2-G05 clone of B-hPSMA MC38 cells was used for in vivo experiments.

 

Post-Inoculation

PSMA expression analysis in harvested B-hPSMA MC38 tumor cells. B-hPSMA MC38 cells were subcutaneously transplanted into humanized B-hPSMA mice. Tumor cells were harvested on day 32 post inoculation and assessed for human PSMA expression by flow cytometry. As shown, human PSMA was highly expressed on the surface of tumor cells, indicating B-hPSMA MC38 cells can be used for in vivo efficacy studies of novel PSMA therapeutics.

Tumor Growth Curve

Subcutaneous homograft tumor growth of B-hPSMA MC38 cells. B-hPSMA MC38 cells (5×105) and wild-type MC38 cells (5×105) were subcutaneously implanted into humanized B-hPSMA mice (male, 8-week-old, n=4 or 5). (A) Average tumor volume ± SEM, and (B) body weight (Mean ± SEM) were measured twice a week. Volume was expressed in mm3 using the formula: V=0.5 × long diameter × short diameter2. As shown in panel A, B-hPSMA MC38 cells were able to establish tumors in vivo, which can be used for efficacy studies.
