Model Selection in Preclinical Studies of Bispecific Antibodies (TCEs): From Tumors to Autoimmune Hotspots

With the increasing number of marketed bispecific T cell engagers (TCEs), research in this field has gained remarkable momentum. TCEs are a class of bispecific antibodies designed to redirect and activate T cells against specific disease targets, offering unique therapeutic potential. While oncology has been the primary focus of TCE development, therapeutic indications are now expanding into autoimmune diseases, bringing new opportunities and challenges. Model selection for pharmacological evaluation plays a critical role in bridging preclinical studies and clinical translation. Current strategies include immune-reconstituted humanized mice combined with tumor models and humanized mice carrying autoimmune disease phenotypes. In oncology, efficacy assessment can be performed using immune-reconstituted mice or humanized mice engrafted with tumor cells. From a translational perspective, humanized models provide robust platforms for pharmacodynamics studies, efficacy validation, and safety evaluation.
