Gene targeting strategy for B-hPDGFD mice.
The human PDGFD CDS and 3’UTR were inserted into exon 1 of mouse Pdgfd gene in B-hPDGFD mice. The promoter and 5’UTR region of the mouse gene are replaced by human counterparts.

Protein expression analysis

Strain specific PDGFD expression analysis in homozygous B-hPDGFD mice by ELISA. Serum were collected from male wild-type mice C57BL/6 mice (+/+) (n=3, 6-week old) and homozygous B-hPDGFD mice (H/H) (n=3, 6-week old), and analyzed by ELISA with human PDGFD ELISA kit. Human PDGFD was exclusively detectable in homozygous mice.

mRNA expression analysis

Strain specific analysis of PDGFD mRNA expression in wild-type C57BL/6 mice and B-hPDGFD mice by RT-PCR. Adrenal gland RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hPDGFD mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human PDGFD primers. Mouse Pdgfd mRNA was only detectable in wild-type mice. Human PDGFD mRNA was exclusively detectable in homozygous B-hPDGFD mice but not in wild-type mice.
