mRNA Expression Analysis

Strain specific analysis of TWEAK and APRIL gene expression in wild type (WT) mice and B-hTWEAK/hAPRIL mice by RT-PCR. Mouse Tweak and April mRNA were detectable only in lung of WT mice (+/+). Human TWEAK and APRIL mRNA was detectable only in homozygous B-hTWEAK/hAPRIL mice (H/H;H/H) but not in WT mice (+/+).

Protein Expression Analysis

Strain specific TWEAK expression analysis in homozygous B-hTWEAK/hAPRIL mice by western blot. Lung was collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTWEAK/hAPRIL mice (H/H), and analyzed by western blot with anti-TWEAK antibody, which has cross reaction between human TWEAK and mouseTWEAK. TWEAK was detectable both in wild-type mice and homozygous B-hTWEAK/hAPRIL mice.
Strain specific APRIL expression analysis in heterozygous B-hTWEAK/hAPRIL mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTWEAK/hAPRIL mice (H/H), and analyzed by flow cytometry with anti-APRIL antibody, which has cross reaction between human APRIL and mouse APRIL. APRIL was detectable both in wild-type mice and homozygous B-hTWEAK/hAPRIL mice.

mRNA expression analysis:

Protein expression analysis:

- TWEAK was detectable in lung of homozygous B-hTWEAK/hAPRIL mice (H/H;H/H) and WT mice (+/+) due to the cross-reactivity of antibodies.
