Protein expression analysis
Western blot analysis of BDKRB2 protein expression in homozygous B-hBDKRB2 mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hBDKRB2 mice (H/H), and then analyzed by western blot with anti-BDKRB2 antibody. 1 μg/μL total proteins were loaded for western blotting analysis by JESS. BDKRB2 was detected in liver and heart in wild-type and homozygous B-hBDKRB2 mice. BDKRB2 was not detectable in brain, lung, spleen, kidney and bladder.
mRNA expression analysis
Strain specific analysis of BDKRB2 mRNA expression in wild-type C57BL/6 mice and B-hBDKRB2 mice by RT-PCR. Heart RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hBDKRB2 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human BDKRB2 primers. Mouse BDKRB2 mRNA was detectable in wild-type C57BL/6. Human BDKRB2 mRNA was exclusively detectable in homozygous B-hBDKRB2 mice.
