Gene targeting strategy for B-hSPP1 mice. The full coding region of mouse Spp1 gene was replaced by the full coding sequence of human SPP1 gene in B-hSPP1 mice.

Strain specific analysis of SPP1 gene expression in wild-type and B-hSPP1 mice by RT-PCR. Mouse Spp1 gene was detectable in splenocytes of wild-type mice (+/+). Human SPP1 gene was detectable only in homozygous B-hSPP1 mice (H/H).

Strain specific SPP1 expression analysis in B-hSPP1 mice by ELISA. Serum was collected from wild-type mice (+/+) and homozygous B-hSPP1 mice (H/H) analyzed by ELISA with species-specific SPP1 ELISA kit. Mouse SPP1 was detectable in wild-type mice. Human SPP1 was exclusively detectable in homozygous B-hSPP1 mice but not in wild-type mice.
