Gene targeting strategy for B-hTIM4 mice. The exons 2-7 of mouse Tim4 gene that encodes the extracellular and transmembrane regions were replaced with human TIM4 exons 2-7 in B-hTIM4 mice.

mRNA expression analysis
Strain specific analysis of TIM4 gene expression in wild-type C57BL/6 mice and B-hTIM4 mice by RT-PCR. Mouse TIM4 mRNA was detectable only in liver of wild-type C57BL/6 mice (+/+). Human TIM4 mRNA was detectable only in homozygous B-hTIM4 mice (H/H), but not in wild-type mice.
Protein expression analysis
Strain specific TIM4 expression analysis in homozygous B-hTIM4 mice by flow cytometry. Macrophages in peritoneal lavage fluid were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTIM4 mice (H/H), and analyzed by flow cytometry with species-specific anti-TIM4 antibody. Mouse TIM4 was detectable in wild-type C57BL/6 mice. Human TIM4 was exclusively detectable in homozygous B-hTIM4 mice but not in wild-type C57BL/6 mice.
